This is fantastic! Vampire Hunter D was one of my early exposures to anime. I tried doing a TMNT hack of Blades in the Dark a while back but I didn't come close to all of the careful consideration you've done here.
Please, please, please at least try to make this happen! Your description of how you would develop the game is incredible! As mentioned below, Paizo was able to create a Vampire Hunter D supplement for 1E, which would imply that the licensing rights could be accessed. Maybe start by asking Paizo how they got permission to make the supplement?
There's a supplement for Pathfinder 1e called "World of Vampire Hunter D", which kinda adapts the material for Pathfinder. Worth checking out. Cheers!

For another take on the genre, there's a very good indie RPG from Voidsprial called 'The Dawnline.' Highly recommended.
